I'd like to know what the difference between these 3 versions of the Gladiator are; is it all down to programming or circuit board changes? Can a MK I be upgraded to one of the other versions, perhaps with a factory kit?

There's no Gladiator MKIII, only Gladiator MKI and Gladiator MKII, which is an improved version of MKI. Gladiator MKII will be out of production soon and will be replaced with Gladiator K, which has the same base with 14 buttons and a mini-throttle, but swappable grips. Gladiator's gymbal is made of plastic.
There's also the current version of the GunFighter MKIII base. This has metal cams (4 sets available), metal springs (5 sets available) and swappable grips.
